THE DIARY GAME || 30th January 2023 || Morning Routine, Laboratory work(Performing gram stain, Observing under microscope and DNA extraction)

Hello, lovely steemians,

I bring you warm greetings from the republic of Ghana which is located in Africa and to be precise west Africa. I am very delighted to come your way today with my daily activities. Just as I always say, it is with great joy that I want to share my daily activities with you and I hope and believe that after going through this entry, you will enjoy it.

Today has being Monday, and it is the first working day of the week. Hence energy from all areas of the work has to come into action. As a result, I woke up as early as 5:45 AM trying to carry out my daily routines as early as possible so that I will be able to report to work before 7 AM. I carried out the cleaning of my room, mobbing it using water, and then I went ahead to get some water for bathing. I sent the water to the bathroom and kept it there. I came back to the room to pick up a sponge and soap after which I went to the bathroom.

I returned to my room and began to dress up. I spent about 9 minutes trying to dress up. After I was done, I got the necessary things I will be needed for the work today. I then got prepared and left the house by putting all my electrical appliances off and getting the keys to lock my door. I locked the door and went out. I spent about ten on my way from the house to the workplace. When I got to the workplace, I first went to log in my details for reporting to work. After I was done, I quickly went ahead to get some documentation done and cleaned my working benches afterward.

When I was done, the service personnel that was around did not come to work today because they have to meet us at an association to elect their leaders in other for them to be helpful in all their activities. Hence when my supervisor reported the work was on the two of us. Hence we decided to group them to help us sort them out in terms of assessing the better needs of all the work. This includes the fact that I will be preparing gram stain so that she will read the slides and the other supervisor will also do the DNA extraction for today.

Because we had agreed on that, it was given to me to start with my portion because we need the gram stain, in other to identify the organisms before we can carry out the prices of extraction. We needed to do that so that we can also finish soon as possible. I quickly picked the slide and labeled them according to the plate IDs. Within a few minutes, I was one carrying out the whole work. After I was done with the work, I noticed the stain reagents are finished. I went to the store to spring some. I then used it to prepare for the slides.

I went ahead to give the slides to my supervisors. Some of the slides were easily identified as the organism we are looking for. Some were also not identified as so. Hence we have to redo the gram stain for those that look like but were not also like. I have to label the slide and did the whole thing for those particular slides again. When I was done, she was able to look at it and gave it a name. According to her, she said it was a different strand of the same organism which means it is not the one of interest to us.

We removed those once and for those that we seem clear, DNA extraction started them. It took several minutes to be able to carry out this great hand work. It took us much high time than expected to finish because we have to wait till the middle in other to be able to finish the extraction well. When we were done, out of the 16 samples, 6 of them were not having enough DNA but the other 10 were good to go with. We then continue disinfecting our working benches and we left. It was around 8 PM. When I got home, I took my bath and rested.
