THE DIARY GAME: FRIDAY, FEBRUARY 03, 2023 (GOING TO THE FARM, AND ACTIVITIES AT WORK)

Hello fellow Steemians,

Guess everyone here is fine? This is a diary of my activities, which serves as a record of my Friday activities both at home and at work side.
Just as I normally do, I woke up at 5:47AM and quickly performed my usual morning chores, after which I took some rice seeds to the farm for them to prepare nursary beds which will be ready for transplantation in about a month time. When I got to the farm, the one to receive the rice and use for the work was already there waiting for me so I greeted him and untied it for him to use.

I sat there for a bit, then he started working, and after a few minutes I jumped on my bike and cycled home because I had to go to work on time. After getting home at 7:25 a.m., I quickly prepared for work by having breakfast and taking a bath before leaving.

I got to work at 8:21AM, so I quickly performed what is required of me every morning before we started working. After this, my first major activity of the day was to stain slides for examination. When my bosses and her colleagues came in, brought out the previous day’s plates for them to physically observe growth. Due to careful observation, some growths were isolated for further examination. They then harvested only a little portion of the organism on the plate and used to prepare slides for microscopic examination. About 25 slides were given to me to stain, so I air-dried them, after which I heat-fixed them before I finally took them to the staining room and stained.

When I was done staining the slides, I then took them with the rack to were the microscope is stationed for observation and analysis.

When further test and careful examination was done, the iDs that qualified to be used for DNA extraction were then isolated and the rest discarded. For all those that passed all the tests, we labeled tubes for them and their colonies were then harvested into them and then used for the extraction. We prepared the volume of reagent required to dissolve the DNA so that we could calculate their concentrations, @edmund.nef was in charge of that. He made sure everything was perfectly prepared as required in the SOP.

Preparing reagents

Whiles the preparation was ongoing, I was sent on an errand to make photocopies and com-binding. I did as I was instructed after which I returned back to work. When I entered there, it was time to take the readings so I joined them and I recorded the results of every sample.

Taking measurements and recording results

We also had some media in the autoclave which was also ready to be brought out and cooled for about 40 minutes before it could be poured into the petri dishes. So when I was done recording all the results, I then went and stored the DNA in their storage box and then kept them in the freezer.

Preparing media
We poured the media into Petri dishes and allowed them to solidify, this actually took less than an hour such that the plates could be turned upside down without the media falling off. After the media solidified, we turned them upside down and labeled them as required and then packed them into their storage refrigerator. All the work benches were then disinfected again after all work and we closed for the day.

I love every aspect of this your entry. You took your time to outline the activities in an orderly manner. I love the fact that you are multi-tasking, which means you are able to do several things at a time.

You alone helping in DNA extraction, quantification, and also preparing media. I love every aspect of it. Keep up the good work.

It is my passion to be doin this molecular work and I hope you will also fellow that pattern if is your passion.
